Functional histological changes of lymphoid organ induced by carrageenan

Abstract
The histological changes of T and B cell areas of lymphoid organs were investigated to study carrageenan (CAR) -induced immunosuppressive effects. Crude CAR(100 mg/kg) was injected intraperitoneally three times into adult male Wistar rats, and they were sacrificed at various intervals to seventh weeks. Changes of peripheral .blood counts and histological examinations of spleen, cervical and mesenteric lymph nodes stained with histochemical method of acid phosphates, a-naphthyl acetate esterase, methyl green pyronin, alcian blue(pH 2.5), and H&E were performed.
The results were obtained as follows:
1. Peripheral blood counts disclosed selective lymphocytopenia, despite of leukocytosis with neutrophilia.
2. Transient atrophic changes of the thymus and mild splenomegaly were observed.
3. In parafollicular areas of the spleen and the lymph nodes, nodular proliferations of CAR-laden macrophages were observed, but there were no evidence of increased acid phosphates activities.
4. In spleen, T cell areas were reduced in size at early times, and restored at third weeks. B cell areas showed transient reduction at early times, but somewhat widened after third weeks.
5. In mesenteric lymph nodes, T cell areas were reduced in size at early times and recovered after fifth weeks. B cell areas were slightly reduced in size.
,6. In cervical lymph nodes, contrast to mesenteric lymph nodes, T cell areas were markedly reduced in size with times, but B cell areas were slightly widened. From the above, immunosuppressive effects of CAR might be induced by functional loss of macrophages as well as reduction of T lymphocytes in lymphoid organ.
